Quality control for lyophilized materials includes visual inspection, residual moisture measurement, and reconstitution testing. Cake appearance can reveal process problems such as collapse, shrinkage, or meltback, although appearance alone does not prove potency. Residual moisture is commonly measured by Karl Fischer titration or by loss on drying. Reconstitution time is checked because a slow or incomplete dissolve can indicate a change in pore structure. Stability studies track these attributes over time under defined temperature and humidity conditions.
Analytical methods for lyophilized solids must account for the low moisture content and the fragile cake. Karl Fischer titration is widely used for water content, while near-infrared spectroscopy can measure moisture non-destructively in sealed containers. X-ray diffraction and modulated differential scanning calorimetry help identify crystalline or amorphous phases. Residual solvent analysis may be needed if organic solvents were used during formulation. The combination of these methods supports batch release and long-term stability assessment.
The physical chemistry of freezing influences whether a formulation forms an amorphous glass or a crystalline solid. Amorphous systems can collapse if product temperature rises above the glass transition temperature of the freeze concentrate. Crystalline systems may show eutectic melting, where ice and solute melt together at a fixed temperature. Formulators add bulking agents, lyoprotectants, and buffers to preserve structure and biological activity. The optimum cycle keeps product temperature below critical thresholds during primary drying while allowing efficient sublimation.
Lyophilization is a dehydration process that removes water from a frozen material by sublimation under low pressure. The material is first frozen so that water becomes ice; then the surrounding pressure is lowered below the vapor pressure of ice. Heat is applied gently so ice changes directly to vapor without passing through a bulk liquid phase. The vapor is collected on a cold condenser, leaving a dry porous matrix. This process differs from simple evaporation because the material remains frozen during the main drying stage.
| Property | Value | Notes |
|---|---|---|
| Storage temperature | 2–8 °C or 20–25 °C | Depends on product stability; some require frozen storage. |
| Moisture content | 0.5–3% w/w | Higher values may reduce stability; target set per product. |
| Moisture method | Karl Fischer titration | Coulometric for low levels; volumetric for higher levels. |
| Cake appearance | Uniform, intact, no collapse | Visual inspection is qualitative and not a potency measure. |
| Reconstitution time | Seconds to several minutes | Depends on cake density, excipients, and diluent. |
Quality control for freeze-dried lots combines visual inspection with instrumental tests. Cake appearance, color, and shrinkage are recorded against a reference, while residual moisture is measured by Karl Fischer titration or loss on drying. Thermal analysis can reveal phase transitions and crystallization events, and X-ray diffraction distinguishes amorphous from crystalline solids. Microbiological tests and container closure integrity checks are also routine for sterile products. Analytical methods must be validated for the matrix, because excipients and low moisture can affect accuracy. Open questions include how best to predict long-term stability from short-term data.
Lyophilized solids are often hygroscopic, so handling occurs in controlled low-humidity areas or glove boxes when the material is exposed. Vials remain sealed with elastomeric stoppers and aluminum crimps until use, because airborne moisture can raise residual water and shorten shelf life. The porous cake is fragile and may crack, shrink, or powder during transport. Personnel typically avoid repeated warming and cooling of sealed units, which can draw moisture through closures. These practices aim to preserve the low water content achieved during drying.
In practice, lyophilization is slower and more energy intensive than simple drying. Cycle times can range from hours to several days depending on load, container, and formulation. Amorphous materials may require excipients that help preserve structure during freezing and drying. The method is widely used for biological materials, pharmaceuticals, and foods where heat drying would cause unacceptable change. Open questions remain about scaling cycles between laboratory and production equipment, and this gap affects technology transfer.
Lyophilization removes water by freezing a material and then lowering pressure so ice changes directly to vapor. The process relies on sublimation, the phase transition from solid to gas without an intermediate liquid state. Because the material remains frozen during primary drying, the structure often stays porous. This porous matrix can rehydrate quickly when water is added back. The low pressure also allows vapor to leave the solid matrix without boiling.
A typical cycle begins with freezing, which fixes the material into a solid and determines ice crystal size. Primary drying then raises heat under vacuum so ice sublimes, often near or below the collapse temperature of the formulation. Secondary drying removes bound water that remains after ice is gone, usually by gently warming the product. Each stage balances heat input against pressure to avoid melting or structural damage. Temperature probes and pressure sensors guide the transition between stages.
Stability of a lyophilized product depends on its glass transition temperature, the temperature at which the amorphous cake transitions from a glassy to a rubbery state. Storage below this temperature minimizes molecular mobility and slows chemical degradation. If the storage temperature exceeds the glass transition temperature, the cake may collapse, shrink, or become sticky. Accelerated stability studies at elevated temperatures and humidity help predict shelf life, but they do not always reflect real-time behavior. Residual moisture content also plays a critical role in long-term stability.
Reconstitution involves adding a suitable diluent, often sterile water or a buffer, to the dried cake. Gentle swirling or inversion helps dissolve the material without creating excessive foam. The time required for complete dissolution can range from seconds to several minutes and depends on the cake structure and the diluent. Improper reconstitution, such as vigorous shaking or using the wrong diluent, can cause protein aggregation or loss of activity. After reconstitution, the product may have a limited shelf life and should be used according to its labeling.
Lyophilized products are typically hygroscopic and require protection from moisture during storage. Manufacturers seal them in glass vials, often under vacuum or an inert gas such as nitrogen. The container closure system, including the stopper and crimp seal, must prevent water vapor ingress. Storage temperature varies from controlled room temperature to refrigerated or frozen conditions, depending on the formulation. Humidity-controlled environments are essential because even brief exposure to ambient air can degrade the product.

=== Immune dysfunction === Those with immunodeficiency (especially infection with HIV) and with neurological disorders that may impact immune system function such as Parkinson's disease (for which the condition is an autonomic sign) and stroke are particularly prone to it.
== Clinical significance == Under diabetic conditions AR converts glucose into sorbitol, which is then converted to fructose. 20466987 It has been found to play an important role in many diabetes complications such as diabetes retinopathy and renopathy. It is also involved in many oxidative stress diseases, cell signal transduction, and cell proliferation process including cardiovascular disorders, sepsis, and cancer. It has been reported that the action of AR contributes to the activation of retinal microglia, suggesting that inhibition of AR may be of a therapeutic importance to reduce inflammation associated with activation of RMG. Adapting AR inhibitors could as well prevent sepsis complications, prevent angiogenesis, ameliorate mild or asymptomatic diabetic cardiovascular autonomic neuropathy and may be a promising strategy for the treatment of endotoxemia and other ROS-induced inflammatory diseases.
=== Election of Commissioners === The Commissioners are appointed into different groups and, initially, for differing terms. Each Commissioner must, at a nominated time, resign and seek re-election at an Annual General Meeting. Commissioners in Group A were required to stand down before the 2013 Annual General Meeting, where they were able to re-apply for their position. Group B members were required to stand down before the 2014 AGM and Group C members were required to stand down before the 2015 meeting. Ten members may, with the additional support of both the NSW and Queensland Rugby Leagues, vote to remove a Commissioner. Fourteen members are required to remove a Commissioner without the support of both the NSW and Queensland Rugby Leagues. Should either of these events occur, the remaining Commissioners would appoint a replacement. A Commissioner who has been removed from office may not seek re-appointment for a period of three years.
The demand for indigo in the 19th century is indicated by the fact that in 1897, 7,000 km2 (2,700 sq mi) were dedicated to the cultivation of indican-producing plants, mainly in India. By comparison, the country of Luxembourg is 2,586 km2 (998 sq mi). In Europe, indigo remained a rare commodity throughout the Middle Ages. A chemically identical dye derived from the woad plant (Isatis tinctoria) was used instead. In the late 15th century, the Portuguese explorer Vasco da Gama discovered a sea route to India. This led to the establishment of direct trade with India, the Spice Islands, China, and Japan. Importers could now avoid the heavy duties imposed by Persian, Levantine, and Greek middlemen and the lengthy and dangerous land routes which had previously been used. Consequently, the importation and use of indigo in Europe rose significantly. Much European indigo from Asia arrived through ports in Portugal, the Netherlands, and England. Many indigo plantations were established by European powers in tropical climates. Spain imported the dye from its colonies in Central and South America, and it was a major crop in Haiti and Jamaica, with much or all of the labor performed by enslaved Africans and African Americans. Indigo plantations also thrived in the Virgin Islands. However, France and Germany outlawed imported indigo in the 16th century to protect the local woad dye industry. In central Europe, indigo resist dyeing is a centuries-old skill that has received UNESCO Intangible Cultural Heritage of Humanity recognition.

=== Neuropsychiatric === Mood disorders like bipolar disorder and clinical depression are the most common conditions underlying catatonia. Other psychiatric conditions that can cause catatonia include schizophrenia and other primary psychotic disorders, autism spectrum disorder, ADHD, and post-traumatic stress disorder. Psychodynamic theorists have historically interpreted catatonia as a psychological defense against the potentially destructive consequences of responsibility, with the passivity of the disorder providing relief.
John Davison Rockefeller III (March 21, 1906 – July 10, 1978) was an American philanthropist. Rockefeller was the eldest son and second child of John D. Rockefeller Jr. and Abby Aldrich Rockefeller as well as a grandson of Standard Oil co-founder John D. Rockefeller. He was engaged in a wide range of philanthropic projects, many of which his family had launched, as well as supporting organizations related to East Asian affairs. Rockefeller was also a major supporter of the Population Council, and the committee that created the Lincoln Center in Manhattan.
== Dietary intake in humans == The European Food Safety Authority (EU) and the US Institute of Medicine, on reviewing existing evidence, have decided there is insufficient evidence to publish a dietary reference value for vitamin K or for K2. They have, however, published an Adequate Intake (AI) for vitamin K, but no value specifically for K2. Parts of the scientific literature, dating back to 1998, suggest that the AI values are based only on the hepatic requirements (i.e. related to the liver). This hypothesis is supported by the fact that the majority of the Western population exhibits a substantial fraction of undercarboxylated extra-hepatic proteins. Thus, complete activation of coagulation factors is satisfied, but there does not seem to be enough vitamin K2 for the carboxylation of osteocalcin in bone and MGP in the vascular system. There is no known toxicity associated with high doses of menaquinones (vitamin K2). Unlike the other fat-soluble vitamins, vitamin K is not stored in any significant quantity in the liver. All data available as of 2017 demonstrate that vitamin K has no adverse effects in healthy subjects. The recommendations for the daily intake of vitamin K, as issued recently by the US Institute of Medicine, also acknowledge the wide safety margin of vitamin K: "a search of the literature revealed no evidence of toxicity associated with the intake of either K1 or K2". Animal models involving rats, if generalisable to humans, show that MK-7 is well tolerated.
=== Journalistic opinions and investigations === Through the years, Ana Leal's investigative journalism in TVI brought new leads to Mendonça's case, always before the police. She followed leads from the Benidorm video, information from various pedophile photos from Holland and Switzerland. According to the journalist "it only has one explanation: negligence from the many teams of the Judiciary Police (...) There wasn't a political desire (..) They got followed TVI's investigations (...) it was annoying for TVI to be ahead (...) shame to a PJ team." In two photos presented in the PJ (Judiciary Police) process for a SIC report, the inspector Luís Bordadágua said that it was possible to see resemblances with Mendonça in the archives found with pedophiles. The reconnaissance was hindered due to the boy having his eyes shut in one of the photos. A Public Ministry agent alleged that the leads were fake and the PJ said later that they did not have leads to follow such a line of investigation. In another photo that Ana Leal had access to in 2002 and gave to the judiciary power, it's possible to see a gagged boy with signs of torture that the family recognized as Mendonça. With the appearance of these leads, Ricardo Sá Fernandes questioned the "worrying social passivity of the authorities of the European Union to destroy the international pedophile ring" and the "lack of capacity for investigation, mainly at a European level".

== Plot == Jesse (Danny McBride), Judy (Edi Patterson) and Kelvin (Adam DeVine) participate in a rehearsal for the "Aimee-Leigh Birthday Celebration Give-A-Thon", hosted by Baby Billy (Walton Goggins). However, during their attempt to lift up, the Gemstones' jet packs malfunction and ruin part of the set. Billy scolds them and demands that they find Eli, who has gone missing for the past few days, as he is vital to the Give-A-Thon's success. Eli (John Goodman) has moved to Florida, living in a yacht where he drinks alcohol and has sex with strangers. Jesse, Judy and Kelvin arrive to get him to take part in the telethon, but Eli refuses, feeling he needs time to consider what he truly wants in life. Nevertheless, they force him to change his mind after repeating the word "pussy" to annoy him. BJ (Tim Baltz) now attends pole dancing classes, although Judy feels jealous of his female colleagues. Kelvin and Keefe (Tony Cavalero) have embraced their relationship and formed a new church program named "Prism", although Keefe wishes they would finally marry or kiss in public. The Gemstones have also launched "Prayer Pods" to the market, but sales are incredibly low. Billy surprises the family by bringing Lori Milsap (Megan Mullally), Aimee-Leigh's long-time musical partner. She and Eli bond, reminiscing over their experiences with Aimee-Leigh. During the telethon, Eli is amazed upon seeing Lori singing, and has a flirtatious encounter with her backstage.
=== Criminal prosecutions and convictions === In 2019, Schmitt launched the SAFE Kit Initiative to reduce the backlog of untested sexual assault kits in Missouri. As of October 2021, approximately two thousand kits had been examined under the initiative and thousands remained to be tested. In January 2020, Schmitt prosecuted a murder case in the City of St. Louis. The jury returned a quick verdict, finding Antonio Muldrew guilty of first-degree murder for shooting and killing Ethiopian refugee Abdulrauf Kadir at a convenience store in 2014. This was the first time a Missouri attorney general prosecuted a murder case in the City of St. Louis. On July 21, 2020, Schmitt filed amicus briefs that argued that "Missouri's statutes specifically authorize Missouri citizens to use firearms to deter assailants and protect themselves, their families, and homes from threatening or violent intruders" and requested dismissal of cases filed by prosecutor Kimberly Gardner against Patricia and Mark Thomas McCloskey for brandishing firearms at protesters who had trespassed on their property while marching in St. Louis in 2020. Schmitt expressed concern about "the chilling effect that this [case] might have with people exercising their Second Amendment rights." Schmitt opposed motions calling for the release of Lamar Johnson, who was convicted for murder on the basis of a single eyewitness's testimony, after a conviction integrity unit found "overwhelming evidence" of Johnson's innocence in 2019. Prior to hearings on his release, Schmitt unsuccessfully called for sanctions on St.
=== Optic neuritis === HHV-6 induced ocular inflammation has been reported three times. All three were reported in elderly individuals, two during 2007 and one during 2011. The first two were reported in Japan and France, the most recent one in Japan. These were believed to have occurred as a result of a reactivation, as anti-HHV-6 IgM antibody levels were low.
dysplasia The abnormal growth or development of a tissue or organ; a change in the growth, behavior, or organization of cells within a tissue, or the presence of cells of an abnormal type, such that the tissue becomes disordered, an event which often precedes the development of cancer.
== Evolution == Most of the aaRSs of a given specificity are evolutionarily closer to one another than to aaRSs of another specificity. However, AsnRS and GlnRS group within AspRS and GluRS, respectively. Most of the aaRSs of a given specificity also belong to a single class. However, there are two distinct versions of the LysRS - one belonging to the class I family and the other belonging to the class II family. The molecular phylogenies of aaRSs are often not consistent with accepted organismal phylogenies. That is, they violate the so-called canonical phylogenetic pattern shown by most other enzymes for the three domains of life - Archaea, Bacteria, and Eukarya. Furthermore, the phylogenies inferred for aaRSs of different amino acids often do not agree with one another. In addition, aaRS paralogs within the same species show a high degree of divergence between them. These are clear indications that horizontal transfer has occurred several times during the evolutionary history of aaRSs. A widespread belief in the evolutionary stability of this superfamily, meaning that every organism has all the aaRSs for their corresponding amino acids, is misconceived. A large-scale genomic analysis on ~2500 prokaryotic genomes showed that many of them miss one or more aaRS genes whereas many genomes have 1 or more paralogs. AlaRS, GlyRS, LeuRS, IleRS and ValRS are the most evolutionarily stable members of the family. GluRS, LysRS and CysRS often have paralogs, whereas AsnRS, GlnRS, PylRS and SepRS are often absent from many genomes.

Most lyophilized products are stored in sealed containers at controlled temperature and humidity. Some require refrigeration, while others are stable at room temperature. Protection from light and moisture is often necessary.
Cake collapse indicates that the porous structure was lost during drying. It can result from excessive product temperature or an unsuitable formulation. Collapsed cakes may have slower reconstitution and are often rejected by visual inspection.
Residual moisture affects the chemical and physical stability of a lyophilized solid. High moisture can promote degradation, aggregation, or cake shrinkage. The acceptable range is set for each product based on stability data.
The main change is sublimation, in which ice becomes water vapor without melting into liquid water. This occurs when the chamber pressure is held below the vapor pressure of ice while mild heat is supplied. The result is a dry, porous solid that retains much of its original shape.
